Description

7-Methyl-6-thioguanosine (MESG, Abs = 330 nm) forms 7-methyl-6-thioguanine (Abs = 355 nm) in the presense of inorganic phosphate and purine nucleoside phosphorylase (PNP) and thus constitutes a rapid and sensitive assay for phosphate. If inorganic pyrophosphatase is present, MESG may also be used to detect inorganic pyrophosphate.

MESG should be used as soon as possible after dissolution.

Description

This nucleoside shows immunosuppressive and antiinflammatory activity, e.g. by inhibiting cellular interactions that result in the lysis of antigen-bearing target cells, a “killer cell” phenomenon thought to play a role in tumor and graft rejection (Krenitsky, T. A.; Rideout, J. L.; Chao, E.; Koszalka, G. W.; Gurney, F.; Crouch, R. C.; Cohn, N. K.; Wolberg, G.; Vinegar, R., J. Med. Chem. 1986, 29, 138-143). When 3-deaza-dA is incorporated into oligonucleotides, the adenine N3 nitrogen is absent in the minor groove, thus providing a useful probe for studies of electrostatic interactions in this region of DNA (Maki, A. S.; Kim, T.; Kool, E. T., Biochemistry, 2004, 43, 1102-1110) as well as how different DNA polymerases use the N3 nitrogen as a substrate specificity determinant (Hendrickson, C. L.; Devine, K. G.; Benner, S. A., Nucleic Acids Res., 2004, 32, 2241-2250).
